Hi Claire, 

As long as this billing method is available to you in your location, you should be able to use a debit/prepaid card to purchase connects for your Upwork account. Billing methods are available based on a user's location. To check if this is available to you, you can go to your Billing Methods page (Settings > Billing Methods), and check which methods are available.
