I recently accepted a job for a new client. The flat fee is $1000. Since Upwork takes 20% up to $500 in lifetime billing per client, does that mean I'll receive $850?

Yes, that's correct. If it's an Enterprise Client or a Featured Job Post, Upwork will take only $100.
