I have applied for many jobs but everyone's demand is 50$ before starting. Can I do it without paying the project fee?

Don't pay anything - they are scams. Clients are supposed to pay freelancers, not the other way round.

Flag those "jobs".

You cannot know in advance whether a scammer will ask you for that absurd fee. One thing you can do to try and filter out some scammers is to check whether the client posting the job has a proven record of work on Upwork (i.e., payments made so far, reviews, etc.).
