I don't want to lose my Upwork account. Please let me know what is the most important rule that needs to follow strictly to prevent permanent account suspension.

Also, don't try to scam buyers. Don't try to coax them to pay you outside of upwork. Don't pressure them to give you high rating. And most important of all, just because you were lucky enough to have an interview but if you don't have the skill to do the work don't accept the job.

But sometimes Upwork suspends an account without leaving any detailed cause of account suspension through email. What is the main cause of that?

Breaking the rules. Just stick to common sense and ToS, and you will be fine.
