Hey guys, I'm new as a freelancer.

So, this month is a tough one because of the holidays and stuff. 

I was wondering if I get paid today can I transfer that money to my Payoneer?

when it should be available the money to spend?

It includes weekends, yes.

So if the client releases the milestone today, you can withdraw to Payoneer on Wednesday.
