Upwork places no limit on the number of Upwork accounts that are used from an individual PC.

Can you and your bother use Upwork from the same computer, using two different Upwork accounts?

Yes.

You already know that Upwork has many automated algorithms and other means for searching for freelancers who are using Upwork inappropriately. If Upwork detects suspicious behavior, they will investigate further. But simply having two users accessing the site from the same computer? That is not a violation of any Upwork ToS.
