Is it possible to to deposit £5000 against my account with upwork?

Later on when I hire free lancers I would like to pay from this deposited money rather then using my card each time.

Unfortunately, that is not possible. You can pay your freelancers using a credit card, PayPal account or bank account. Please, check this help article for more information.
