Simple answer: When you hire the freelancer.
Longer answer: Upwork uses a true escrow system.
If you hire John to do a project for $100, your credit card is charged immediately. But the money doesn't go to John yet. The money is in an escrow account. When John sends you the work, you review the work and then click a button to release that money from the escrow account to him.

The charge happens when you send an offer.
This is important because if you send an offer, and end up taking it back because the worker didn't respond, and you have to withdraw that offer to send to another worker, you'll be charged again.
Of course, you'll get that first charge back in a few days. Just something to be aware of.
