If this is a fixed-price contract:

1) Do the work

2) Submit it using the "Submit Work / Request Payment" button attached to the contract listing.

I'm not sure if you are referring to a specific job, or submitting work on the platform. For hourly contracts, you and your client can choose which method is best for both you when submitting work on a project. Make sure you use the Upwork Desktop App to log time for your project, and note appropriate memos when logging time. You may refer to this help article fo rmore information. 

For fixed-price contracts, please click on the Submit Work/Request Payment button to submit work on a project. You may read more about it here.
